利用透明颤菌血红蛋白基因在谷氨酸棒杆菌中的表达提高谷氨酸和谷氨酰胺的产量

Improving Production of Glutamic Acid and Glutamine Using Recombinant Corynebacterium glutamicum Harboring Hemoglobin Gene from Vitreoscilla sp.

  • 摘要: 为了降低谷氨酰胺 (glutamine,Gln)生产过程中所需要的高溶氧水平对设备的要求 ,在谷氨酸棒杆菌 C.glutamicum ATCC14 0 6 7中表达增强摄氧的透明颤菌血红蛋白基因 (Vitreoscilla hemoglobin gene,vgb) ,以此提高细胞的摄氧能力及能量的供给水平 .对得到的重组菌和野生菌进行了发酵实验 .在溶氧只有 5 %的条件下 ,重组菌比野生菌细胞干重提高了 1.2倍 ,单位细胞谷氨酸的生产能力提高了 6 .5倍 ,单位细胞谷氨酰胺的生产能力提高了 1.4倍

     

    Abstract: The expression of Vitreoscilla hemoglobin gene (vgb gene) in Corynebacterium glutamicum ATCC 14067 increased the oxygen and energy supply resulting in improving glutamic acid (Glu) and glutamine (Gln) productivity. Under 5% dissolved oxygen, the cell dry weight (CDW) of the recombinant strain and wild strain were 45.1 g/L and 54.1 g/L respectively, the concentration of Glu were 9.56 g/L and 1.27 g/L, the concentration of Gln were 5.51 g/L and 2.27 g/L. The results indicated that increasing oxygen supply via vgb gene expressing in recombinant C. glutamicum remarkably promote cell growth and production of Glu and Gln. This could be an effective way to reduce the cost of industrial fermentation of Glu and Gln.
